Protocol for intradermal injection
Intradermal injection(ID) is a shallow or superficial injection of a substance into the dermis, which is located between the epidermis and the hypodermis. Which has the longest absorption time of all parenteral routes and has been applied in many aspects such as allergy and local anesthesia tests. In this protocol, we here present the experimental procedure of ID in the mouse as follows.
STEP 1
Restrain and anesthetize the animal before surgery. Use clippers to shave or pluck an injection site on the back of the animal to remove the hair, then swab the site with 70% ethanol.
STEP 2
Insert an appropriately sized needle into the skin at an angle of 15-30° until there exists the resistance.
Alternative method: gently pull the skin near the injection site taut and insert the needle at an angle nearly parallel to the plane of the skin. Which is useful in mice because it can prevent them from moving during the injection.
STEP 3
A bleb, which has a light color than the surrounding skin, will appear at the injection site when needle placement is correct. After injection, gently apply pressure to prevent the solution reflux.
STEP 4
Place the mouse back to it’s cage next to the electric heater until it recovers.
