Introduce
This novel transfection enhancer (Enhancer A+B) is composed of liquid A (1000×) and liquid B (200×). It contains high-valent cationic polymer and cholesterol, which can enhance the permeability of the membrane and significantly improve the infection efficiency of retrovirus on cells. When infecting cells with lentivirus, adding Enhancer A+B can increase the infection efficiency of lentivirus by 4-10 times, which is better than polybrene. For difficult-to-transfect cells with lentivirus, the use of transfection enhancer can make viral infections on cells easier, which will largely contribute to subsequent experiments.
Procedure
1. Cells seeding
Each well (Taking a 6-well plate as an example) is plated with 1×10
6 cells to be infected and cultured with 2 mL of complete medium per well.
2. Viral infection
24h after cell inoculation, replace with fresh complete medium. The optimal MOI of the cell can refer to the literature (or determine the optimal MOI of infection by virus MOI gradient experiment). Calculate the amount of virus used, add the corresponding volume of lentivirus suspension for infection, then gently shake the six-well plate to make it evenly mixed.
3. Add transfection enhancer
Immediately after inoculating the virus suspension, add the transfection enhancer. Taking 2 mL of culture medium as an example, add 2 μL of liquid A and 10 μL of liquid B, gently shake the six-well plate to make it evenly mixed.
(This product can be selected according to the difficulty of virus infection, and stored at -20°C after use)
4. Change media
After 4-6 hours post infection, remove the virus supernatant and replace with fresh complete medium.
5. Fluorescence detection
The fluorescence effect of virus infection was observed under a fluorescence microscope 48-72 h post infection.

Note: The novel transfection enhancer showed no obvious cytotoxicity for most cells by testing, but there still exists the possibility that a small number of cells are sensitive to it. If the cell state is significantly deteriorated or lysed and died after adding the novel transfection enhancer, which means that this product is not applicable to this kind of cells. Therefore, the transfection enhancer should be removed and the cells should be infected with lentivirus suspension alone.
