Protocol for subcutaneous injection
A subcutaneous injection is a shot given into the fat layer between the skin and muscle. Subcutaneous injections are used to give small amounts and certain kinds of substance. In this protocol, we present the experimental procedure of the subcutaneous injection in the mouse as follows.
STEP 1
Manually restrain the animal using the scruff method to allow access to an area with loose skin and to creat a subcutaneous space.
STEP 2
With the needle of appropriate size pointing away from you, insert at a 30°- 45° angle in the subcutaneous space, being careful to not push through the other side.
STEP 3
Aspirate the syringe to ensure negative pressure prior to injecting. If blood is drawn back into the hub of the needle, then withdraw needle and repeat placement. If there is negative pressure, proceed with the injection and depress the plunger until the solution has been fully administered.
STEP 4
Remove needle when complete the injection, gently apply pressure to prevent backflow of the solution.
STEP 5
Return the mouse to its cage and take good care of it.
